Determining Chromosome Counts

OK. Here's a question for those with formal Biology training:

What equipment is necessary to determine chromosome count?

I have access to high power (1000x) optical microscopes at work. Is
this sufficient?

How is the sample preparation done?

What stains are used?

It would be fun to do counts on my plants to help guide hybridization
attempts...
