I think Dave doesn't want a chromosome counting protocol, but exactly
extracting DNA, I think for DNA-research, possibly involving PCR.
I am posting the method to him personally, if anyone else is interested, mail
to me personally, because I think this is too far off-topic for the whole
list.
BTW, if somebody is interested in a protocol for making chromosome
preparations, I can also supply two different methods to everyone who is
interested.
